# How tirzepatide works: GIP and GLP-1, explained

Tirzepatide acts on two hormone receptors involved in appetite and glucose regulation. Here is what that means in plain English.

Tirzepatide activates two types of receptors: GIP and GLP-1. These receptors respond to signals involved in appetite and blood glucose regulation. The medication is sometimes described as having a dual action because it works through both.[1]

That describes its biology. It does not mean twice the weight loss or twice the benefit for every person.

## What the two signals do

GIP stands for glucose-dependent insulinotropic polypeptide. GLP-1 stands for glucagon-like peptide-1. Both are hormones the body makes, and tirzepatide is designed to activate their receptors.[1]

According to the approved prescribing information, tirzepatide increases insulin secretion in a glucose-dependent way, reduces glucagon secretion, and improves insulin sensitivity. In simpler terms, it affects how the body responds to and manages glucose.[1]

It also reduces calorie intake through effects on appetite. Its action in weight management therefore includes an effect on appetite.[1]

## Where digestion fits

Tirzepatide delays stomach emptying. That effect is largest after the first dose and diminishes over time.[1] It is one part of the medication's action, not a complete explanation for every change in hunger, fullness, or digestion.

Nausea, vomiting, diarrhea, and constipation are among the common adverse reactions.[1] Stronger digestive symptoms are not evidence that someone is getting a better result.

## What the mechanism cannot predict

Knowing the receptor targets cannot tell an individual how much weight they will lose, how quickly it will happen, or whether they will tolerate the medication. Those questions require evaluation and follow-up.

Research and prescribing information for an FDA-approved tirzepatide product also do not establish that a compounded formulation has the same safety, effectiveness, or quality. Compounded drugs are not FDA-approved.[2]
