# Semaglutide vs. tirzepatide: what single and dual action mean

Semaglutide targets GLP-1 receptors; tirzepatide targets GIP and GLP-1 receptors. How that difference fits into a treatment comparison.

Semaglutide activates GLP-1 receptors. Tirzepatide activates both GIP and GLP-1 receptors. “Dual action” describes that receptor activity; it does not mean the medicine is twice as effective or automatically the better choice. [1][2]

A meaningful comparison also includes the condition being treated, the exact product, side effects, other medicines and the person's medical history.

## How the signals differ

GLP-1 is involved in appetite and glucose regulation. Semaglutide's effects include reduced calorie intake and glucose-dependent effects on insulin and glucagon, hormones involved in blood glucose control. It also slows stomach emptying. [1]

Tirzepatide acts at GIP receptors as well as GLP-1 receptors. Its label describes effects on appetite, insulin secretion, glucagon and insulin sensitivity. Receptor activity explains how the medicine works; it is not a complete prediction of an individual's response. [2]

## Comparing outcomes requires comparable evidence

Results from separate trials can involve different populations, treatment durations and study designs. Those differences matter when reading a headline about weight change. A study average is not a forecast for one person.

The product studied also matters. Evidence from an FDA-approved product does not make a compounded preparation FDA-approved or interchangeable with it. FDA does not review compounded drugs for safety, effectiveness or quality before marketing. [3]

## Safety can decide the choice

Both medicines have contraindications and warnings, including specific thyroid-tumor history, serious allergic reactions and gastrointestinal risks. Pregnancy plans and other medicines must also be considered. Tirzepatide's labeling includes additional precautions about oral hormonal contraception. [1][2]

The decision therefore cannot be reduced to the number of receptors. A useful comparison explains why a particular option is being considered, what will be monitored and how problems will be handled.
