# NAD+ and NMN: what human research actually measures

Raising an NAD-related blood marker is different from improving fatigue or extending life. Human trials answer narrower questions than many aging claims.

NAD+ is involved in essential cellular processes, including metabolism. NMN is one compound the body can use in NAD+ production. That biology has prompted research, but it does not by itself show that a supplement or injection improves everyday energy or extends human life. [1]

The key is to look at the outcome each human study actually measured.

## A higher marker is one kind of finding

A small randomized crossover trial of nicotinamide riboside, another NAD+ precursor, found that it increased measures of NAD+ metabolism in healthy middle-aged and older adults. The study was designed around that precursor, not an injectable NAD+ treatment for fatigue. [2]

A separate 60-day trial in 80 healthy middle-aged adults studied NMN. It reported increases in blood NAD concentration and better results on a six-minute walking test in treated groups compared with placebo. No safety issues were identified during the trial. [3]

Those are findings from a short study in a specific population. A walking test and a blood marker are different from years of follow-up measuring illness, disability or lifespan. [3]

## What does “biological age” add?

The NMN study also used a calculated biological-age measure. A change in a calculated score is an outcome of that particular measurement method; it is not observation that someone lived longer or reversed aging. [3]

## Product and route are part of the evidence

An oral precursor study cannot automatically establish the effects of an NAD+ injection. The ingredient, route, participants and duration need to match the claim being made. [1][2][3]

The broader NAD+ review describes unresolved questions about safely translating this biology into benefits for human aging. [1]

For an energy claim, the useful question is whether a trial measured the fatigue problem you care about. “Raises NAD” and “helps me function better” are different claims, and the evidence should say which one it supports.
